Myc tag fusion protein - detection, blocking

Myc Tag is derived from the c-myc gene expression product and its sequence is EQKLISEEDL. Myc Tag monoclonal antibody (clone 2D5), known for its high affinity, detects natural and denatured Myc fusion proteins and is widely used in WB, IF, and IP experiments. Under natural elution conditions, the Myc-tagged polypeptide is used to compete with the recombinant protein to gently elute the Myc-tagged protein.

A mouse anti-c-myc tag antibody was developed in 1985 and used as an immunochemical reagent in the fields of cell biology and protein engineering. The Myc tag is equivalent to the human c-Myc 410-419 peptide (sequence: EQKLISEEDL). The Myc tag can be placed at the C-terminus or the N-terminus, but the low pH elution conditions of the Myc recombinant protein tend to reduce protein viability, so the Myc tag system is widely used for detection but is rarely used for purification. The Myc tag has been successfully used in WB hybridization, immunoprecipitation, and flow cytometry. It can therefore be used to detect the expression of recombinant proteins in bacteria, yeast, insect cells and mammalian cells.
