ProcartaPlex immunoassay common problems and solutions

eBioscience 's Luminex Liquid Phase Chip Detection Reagent - Procarta Plex provides a variety of multi-factor detection protocols for your biological research, including cytokines, growth factors, chemokines and other proteins. There are many problems when selecting the kit and operation. The common problems and answers are summarized below for your reference:

If you accidentally keep the entire kit at -20 ° C. Can this kit be reused?

I accidentally stored the whole kit at -20°C. Can I use it anyway?

A. In addition to the microspheres in the kit, the rest can be stored at -20oC . If the microspheres are stored below 0oC , the microspheres will be damaged and the microspheres will no longer be used.

All the components included in the ProcartaPlex kit are viable except the bead mixture. Storage of the beads at temperatures below 0oC will damage the beads and render them unusable.

If I have purchased a multi-factor kit and a single-factor kit at the same time, do I need to buy the basic kit again?

Do I need to purchase a Basic Kit when combining simplex Kits with a Multiplex Kit?

A. No.

No, when combining a Multiplex kit with Simplex Bead Sets, the Basic Kit is not required. All the non target specific reagents which are included in the Basic kit are components of our Multiplex kits. Only when you are combining multiple Simplex kits (ie no Multiplex kit in the panel) will you need to also purchase the Basic Kit.

Can I use the Mouse Basic Kit with a human single factor test kit?

Can I use the ProcartaPlex Mouse Basic Kit with ProcartaPlex Human Simplex and vice versa?

A. can not. The basic kit is species specific. Basic kits of different species cannot be used interchangeably.

No, the Basic Kits are specific to the species listed, ie the Mouse Basic Kit can only be used with Mouse Simplex Bead Sets. The Mouse, Human, Canine, Porcine, Rat and NHP ProcartaPlex Basic Kits are not interchangeable.

Multi-factor test kits are not purchased at the same time. Can they be used together?

Can I combine ProcartaPlex kits, which were not purchased on the same order? For example: I purchased a kit 3 months ago and another kit today. Can I combine them or do the kits have to be shipped on the same order?

A. Yes. As long as the microsphere number of the kit for simultaneous detection is not repeated, the pre-sample processing is the same.

Yes, you can combine ProcartaPlex kits from different orders to run a bigger multiplex experiment. You will want to ensure that there is no overlap of bead regions between the kits you areituated. Please refer to the online ProcartaPlex Panel Configurator.

Can Luminex 's microspheres be detected on a normal flow cytometer?

Is it possible to measure Luminex beads on a flow cytometer?

A. No. Detection ProcartaPlex (Luminex) can use Luminex 100/200, MAGPIX, FLEXMAP 3D.

To analyze ProcartaPlex (Luminex) plates, you will need access to a Luminex machine such as a Luminex 100/200, MAGPIX, FLEXMAP 3D.

How many microspheres are recommended for Luminex testing?

How many beads do you recommend we collect? And how do we setup the Luminex machine?

A. It is recommended to collect 50-100 each microsphere .

Please collect 50-100 beads per bead region as stated in the product manual.

Can the ProcartaPlex ELISA plate be used to detect the plate on multiple occasions without loss of signal?

Can a finished ProcartaPlex plate be read multiple times on the Luminex instrument without a significant signal loss?

A. Yes.

Yes, it is possible to reread a finished ProcartaPlex plate without a loss in the signal or the number of beads counted. Please note that the Luminex instrument adds additional liquid to the wells with each analysis. It is possible that the wells may become overfilled with Fluid after the third analysis. Hence, we do not recommend reading the ProcartaPlex plates more than two times.

If the ProcartaPlex operation is completed, it cannot be detected in time. So how can I save?

I have completed the ProcartaPlex assay but can't run my samples immediately. How long can I store the ProcartaPlex plate before analysis on a Luminex instrument without a significant loss of signal?

A. Normally, we recommend to check on the machine immediately; if you need to save, it is recommended to save at 2-8oC . But the time cannot exceed one day.

Ideally, we recommend that you analyze the samples immediately. If you do need to store your samples, overnight storage at 2-8°C shows no significant signal loss. We do not recommend storing the ProcartaPlex plate later than 1 day.

Washing step, can I use the centrifugal method?

Can I use a centrifuge for the washing steps in the assay?

A. No.

No, we do not recommend centrifuging the magnetic beads for prolonged periods of time.

After the standard in the kit is dissolved, can it be stored again?

Can I store reconstituted standard protein for later usage?

A. Not recommended. It is recommended to use new standards for each test.

The recombinant standards included with the ProcartaPlex assay are one-time use standards. After reconstituted for use, we do not recommend storing and reusing this recombinant standard. With each new experiment, we recommend reconstituting a fresh unused vial of recombinant standard protein.

Where is the initial dissolved mother liquor concentration of the standard?

Where can I find the stock concentration of the recombinant standards?

A. Please find it in the instructions received with the goods.

This information can be found on the Certificate of Analysis provided with the ProcartaPlex assay.

In the experiment, can the standard curve and the sample be tested for single hole?

When preparing standards/samples, can I run them as single points?

A. For better accuracy of results, we recommend at least duplicate holes in the sample and standard curve wells.

For better accuracy, we recommend that you run samples/standards as duplicates at a minimum.

If I have to stop the experiment, where can I stop?

I have to cut short the assay and can't complete the assay. At which step can we incubate the plates overnight with affecting the assay?

A. Follow the instructions and you can react overnight after adding the standard and sample.

As stated in the product manual, after the addition of the standards and samples, the plate can be stored overnight (Step 11.1.5 or 11.2.5).
