Huantian Granules is the experience of the inherited insomnia of Liu Yongnian, the instructor of the national old Chinese medicine expert's academic experience. Therefore, the rule is to relieve the liver, to promote blood and soothe the nerves. The role of Huantian Granules in the sedation and hypnosis of mice is now discussed.

1 Experimental materials

From the preparation room of Nanjing Traditional Chinese Medicine Hospital, Chinese herbal medicines such as Acacia, Jingtian Sanqi, Mudanpi, Oyster, and Yuanzhi were made into Huantian granules. The daily dosage of adult 30g (formulation amount) ö60kg, batch number: 040406. Kunming (ICR) mice, weighing 18-22 g, male and female, were supplied by the Experimental Animal Center of Nanjing University of Traditional Chinese Medicine. Certificate of Compliance: SYX K (Su) 2002 - 0053. Before and after administration, the experimental mice were divided into male and female cages and fed with full-price extract feed (provided by Jiangsu Synergy Medicine Biotech Co., Ltd.), free drinking water, room temperature: (20 ± 2) ° C, humidity: 50% to 70%.

2 Methods and results

211 Test method

(1) Effect on the number of mice falling asleep: 60 ICR mice, male and female, were randomly divided into 5 groups, namely 1 blank control group: normal saline 2m l; 2 Huantian granule low dose group: 215gökg 3 Huantian granules in the middle dose group: 5gökg; 4 Huantian granules high dose group: 10gökg; 5 stable positive control group: 50m gökg. Each group was intragastrically administered in a volume of 011 m lö10 g for 3 consecutive days. After 30 minutes of the last administration, each group was intraperitoneally injected with a sub-threshold sleep dose of 25 m gökg of pentobarbital sodium. The number of mice that fell asleep in each group within 30 minutes of sodium pentobarbital injection was observed and recorded (as the sleep-reversing reaction disappeared for more than 1 minute, as a sleep-sleeping reaction). (2) 50 ICR mice were randomly divided into 5 groups, each group was male and female, and the specific grouping method was the same as 211(1). Each group was intragastrically administered in a volume of 011 m lö10 g for 3 consecutive days, and then each index was observed before administration and 3 days after administration. The experiment was started at 9:00 am. Observe and record the general behavior of the mice: including posture, gait, presence or absence of drooling, muscle twitching, etc. The mice were recorded spontaneously within 5 minutes of each phase after administration by a small animal analyzer.

(1) Oral Huantian Granules 5gökg and 10gökg can increase the sleep rate of the sub-threshold sleep dose of pentobarbital sodium mice, which is significantly different from the blank control group (P < 0105 or P < 0101). ). It is suggested that Huantian Granule can synergize with the sleep of the subthreshold sleep dose of pentobarbital sodium mice, and has a sleep-inducing effect. (2) Huantian Granules can reduce the spontaneous release of mice by oral administration of 215gökg, 5gökg and 10gökg, which is significantly different from the number of spontaneous activities in the blank control group (P < 0105), 3 dose groups. No symptoms such as salivation and muscle twitch were observed after administration.

3 Discussion

Huantian Granules is a combination of Acacia and Sedum, and it has the effect of reaching Yu'an God, promoting blood circulation and removing blood stasis. Fangzhi's Polygala has calming and anticonvulsant effects. Corundum itself has central inhibition effects such as sedative, analgesic and antispasmodic. One of the main volatile components, paeonol, has analgesic, sedative, hypnotic, anti-inflammatory, antipyretic and anti-allergic effects. Enhance immune function and other functions. This study shows that the high dose of Huantian Granules has a better hypnotic effect on mice. We believe that this may be the synergistic effect of the various drugs in Huantian Granules. In addition, the amount of Acacia skin in the high-dose Huantian Granules It did not reach the dose achieved by insomnia caused by large doses of Acacia skin in the relevant study. Studies have shown that Huantian Granules have no obvious acute toxic effects, and can increase the number of sleeps in pentobarbital sodium mice with subthreshold sleep dose, reduce spontaneous activity of mice, and have sedative and hypnotic effects.
