1. Close all water supply lines and air lines. Open the steam line valve. At the same time, the exhaust valve of the fermenter jacket is slightly opened, and the remaining water of the jacket is discharged.
2. Turn on the fermenter stirring motor and rotate to 200 rpm to heat the fermentation broth. When the temperature rises above 95 °C, the stirring can be stopped. Then, when the temperature rises to 121 ° C (the tank pressure is 0.1 to 0.12 MPa), the timing can be started.
3. When the timing starts, the fermentation time is generally 20-30 minutes. During this time, the temperature should not be lower than 120 ° C. At the same time, the air filter and air pipe can be sterilized.
4. Sterilization of air filter and air duct: slightly open the drain valve of the filter and the diaphragm valve of the air duct to ensure steam sterilization of the air duct. But it can't be opened too much, so as to avoid a large amount of steam entering the tank, and dilute the medium.
5. The steam valve and outlet valve of the discharge and sampling valves are slightly opened to ensure the sterilization of the pipeline. The inoculation port on the lid of the fermenter also needs to be deflated to meet the sterilization requirements.
6. When the insulation is over, the diaphragm valve in the air line should be closed first. Close the air filter drain valve and close the sampling valve outlet valve and the inoculum nut. Then close each steam valve.
7. Open the cooling water valve and drain valve, and open the air flow meter and air vent valve to dry the air filter. At this point, attention must be paid to the change in tank pressure. Never allow the tank pressure to be below 0.02Mpa. And when the tank pressure reaches 0.05Mpa, immediately open the air line to ensure that the tank pressure of the fermenter is about 0.05Mpa.
8. When the temperature drops to 95 ° C, the agitation can be turned on. When the temperature is lower than 50 °C, the automatic temperature control state can be cut, the medium is brought to the inoculation temperature, and the sterilization process is finished.

Second, the operation of the fermentation process
1. Inoculation: The inoculation method may be a flame inoculation method or a differential pressure inoculation method.
(1) Flame inoculation method: Disinfect the inoculation port with an alcohol ring, then open the inoculation cap and quickly pour the inoculum into the can and tighten the cap.
(2) Differential pressure method: Put the gasket before sterilization, open the inoculation cap when inoculation, and pour a certain amount of alcohol into the disinfection first. After a while, insert the needle of the seed bottle into the gasket of the inoculating port. Using the pressure inside the tank and the pressure difference in the liquid bottle, the seed liquid is introduced into the tank and the lid is tightened.
2. Tank pressure The tank pressure must be manually controlled during the fermentation process, that is, the pressure in the tank is controlled by the outlet valve. To adjust the air flow, the outlet valve must be adjusted at the same time, and the pressure inside the tank should be kept constant greater than 0.03Mpa.
3. Measurement and Control of Dissolved Oxygen (DO) (1) Calibration of Dissolved Oxygen: The dissolved oxygen DO value at the constant fermentation temperature and the amount of dissolved air and the maximum amount of air was taken as 100% before inoculation.
(2) Dissolved oxygen DO measurement and control during fermentation: DO control can be achieved by adjusting the air flow rate and adjusting the rotational speed. The simplest is the associated control of speed and dissolved oxygen. Secondly, the intake air amount (manual) control must be adjusted at the same time. Sometimes it is necessary to pass pure oxygen (as in high-density culture of certain genetically engineered bacteria) to achieve the required DO value.
4. Measurement and control of pH (1) Correction of pH: The pH of the PH electrode should be corrected before sterilization.
(2) The pH value is controlled during the fermentation process by using acid and alkali in the peristaltic pump. The acid or alkali bottle must be sterilized in a sterilizer.

Third, the operation of the controller
1. Startup of the controller: Turn on the power, first press the “S/E” button on the membrane keyboard, and then press the “confirm” button, the fermentation control program starts; at this time, if the water in the heater is not full, the program The water inlet operation will be automatically performed; after the water is full, the user can control each actuator according to the above-mentioned operation method of the lower machine controller.
2. Operation of the controller: Use the F1~F6 buttons to switch the interface in the LCD screen to the interface that the user needs to control. Use the arrow keys to move the cursor in the interface to the variable to be controlled. If the operating mode is changed, directly Press the Enter key. If you need to enter a number on the keyboard, press the Enter key after entering the number. Such as:
(1) Temperature control: In the manual mode, manual operation of the temperature is relatively simple, and only the control amount of the manual state needs to be changed. Enter the temperature control interface by selecting the shortcut keys (F1 ~ F5), then move the cursor to point it to "manual mode", press the "confirm" button, then enter the manual mode of temperature control. At this point, a small hand appears after the "Manual Mode" to indicate that the current selection is manual. Fourth, the operation of the steam generator
1. Open the water inlet switch to keep the water level to the highest level and keep it in the water.
2. Connect the generator power supply and supply water to the normal water level (50-80% of the liquid level tube). Do not exceed the maximum water level, and must not lower than the lowest water level. Close the steam outlet valve.
3. Plug in the power, turn on the power switch, the work indicator lights up, start to heat the pot water
4. Connect the steam tube to the fermenter sandwich line system, open the relevant valve, keep the line open, and close the fermenter outlet valve. When the pressure rises to the working pressure (2kg/cm2), the steam outlet valve is opened, that is, the steam is available.
5. After use, first turn off the power, then close the inlet pipe. After the generator is properly cooled, drain the sewage from the pot.
【Precautions】
1. The setting pressure of the safety valve has been adjusted by the manufacturer and should not be adjusted at will. If the safety valve is found to be malfunctioning, the new safety valve should be replaced. It is strictly forbidden to change the automatic pressure control function and parameters.
2. During normal operation, discharge the oil at least once every 8 hours, and timely descale the water level probe with sandpaper.
3. It is strictly forbidden to close the safety valve during use. It is strictly forbidden to modify it or block it with a plug.
4. The pressure gauge trap should be removed and cleaned regularly.

Fifth, the operation of the air compressor
1. Plug in the air compressor and turn on the air compressor to start the machine for 15 minutes without load.
2. If there is no abnormal sound after starting, close the air outlet, and connect the air compressor outlet pipe to the air purifier, and connect it to the fermenter air intake pipe.
3. When the air pressure rises to 2kg/cm2, open the air supply valve, turn on the air switch, and supply sterile air to the sterilized fermenter. After the air pressure reaches the set pressure, the pressure switch automatically cuts off the power and the motor stops running.
4. The operating pressure of the air compressor must not be higher than the rated working pressure. If adjustment is required, it must be carried out by a special salesman.

Sixth, maintenance and maintenance

1, precision filter, the general use period is one year. If the filtration resistance is too large or the filtration capacity is lost, which may affect normal production, it needs to be cleaned or replaced (recommended for direct replacement, no cleaning, and the performance of the filter cannot be reliably guaranteed after the cleaning operation).
2. When cleaning the fermenter, please use a soft brush to scrub, do not use a hard scraper to avoid damage to the surface of the fermenter.
3. The supporting instruments should be verified once a year to ensure normal use.
4. Electrical equipment such as electrical appliances, instruments, sensors, etc. are strictly prohibited from directly contacting water and steam to prevent moisture.
5. When the equipment is stopped, it should be cleaned in time, draining the residual water in the fermenter and each pipe; loosen the fermenter lid and hand hole screws to prevent permanent deformation of the seal.
6. Carbon steel equipment such as operating platform and constant temperature water tank should be painted regularly (once a year) to prevent corrosion.
7. Always check the oil level of the reducer. If the oil is not enough, it needs to be increased in time.
8. Regularly replace the reducer lubricant to extend its service life.
9. If the fermenter is not used temporarily, the fermenter should be empty and drained from the tank and the remaining water in each pipe.

Seven, matters needing attention

1. It must be ensured that all single-piece devices can be used in normal operation.
2. When the filter is disinfected, the steam pressure flowing through the air filter must not exceed 0.17 MPa, otherwise the filter element will be damaged and the filtering capacity will be lost.
3. During the fermentation process, ensure that the tank pressure does not exceed 0.17 MPa.
4. In the actual elimination process, when the jacket is preheated by steam, the inlet steam pressure must be controlled within the working pressure range of the equipment (not exceeding 0.2 MPa), otherwise the fermentation tank may be damaged.
5. When emptying and realizing, be sure to drain the remaining water in the fermenter jacket. Otherwise, the inner cylinder of the fermenter may be flattened, causing damage to the equipment; in the case of actual elimination, too much condensed water may cause the culture solution to be diluted, thereby failing to meet the process requirements.
6. During the cooling process after the end of the air consumption and the end of the actual elimination, it is strictly forbidden to generate negative pressure in the fermenter to avoid pollution and even damage the equipment.
7. During the fermentation process, the tank pressure should be maintained between 0.03 and 0.05 MPa to avoid pollution.
8. During each operation, the pressure in the air pipe must be kept larger than the tank pressure of the fermenter. Otherwise, the liquid in the fermenter will flow back into the filter, block the filter element or invalidate the filter.
9. If you encounter problems that you can't solve, please contact the company's after-sales service department directly. Do not forcefully disassemble or repair.
